Dissolution UV-online Automatic Detection System
Dissolution UV-online Automatic Detection System (RT600-UV) is the latest product of Raytor Instruments based on the third generation dissolution operating system. RT600-UV system includes Automatic Dissolution Apparatus, Automatic Sampling Workstation and Ultraviolet-Visible Spectrophotometer. RT600-UV supports automatic dissolution sampling and on-ine ultraviolet detection. It is more convenient for users to develop drugs and routine detection, and greatly improves work efficiency
Each dissolution vessel has individual flow cell and pipe, which effectively avoids crosscontamination between samples from different dissolution vessel
Automatic dissolution apparatus, automatic sampling workstation and the ultraviolet-visible spectrophotometer are controlled by the same dissolution operating system in the automatic dissolution apparatus. It supports the preset and invokes the on-line ultraviolet automatic detection method for dissolution. The dissolution system will automatically complete the automatic dissolution sampling and the automatic on-line ultraviolet detection according to the preset method. The test results are fed back to the interface of the dissolution operating system in real time, and the protected electronic data documents are automatically generated in real time.
Advanced dissolution operating system can ensure that it fully meets the requirements of data integrity, audit track, account management and other regulatory requirements.
Excellent Accuracy
Thanks to the advanced automatic sampling technology of Instruments, the precision of Dissolution UV-online Automatic Detection System can reach a high level. Excellent accuracy can ensure that the on-line ultraviolet test can truly reflect the real-time dissolution of samples, and the test results are more reliable.
Test cases:
Continuous detect the absorbance of salicylic acid solution (the concentration of salicylic acid was 6, 9, 12, 18, 24 and 36 ug/ml) by manual sampling and ultraviolet on-line respectively. Linear regression curves were established with salicylic acid concentration as X-axis and absorbance (Abs) as Y-axis, and calculate the R-square values of each curveThe results of on-line ultraviolet test showed excellent linear correlation, R-square value was 0.9995, ano there was no significant difference with the manual sampling control group.
Linear regression curves were established with the absorbance (Abs) of the manual sampling control group was X-axis and the absorbance (Abs) of UV-online test group was Y-axis. The R-square value was 0.9996. The results show that Dissolution UV-online Automatic Detection System has excellent accuracy.
Stable Test Results
The test results need not only accuracy, but also stability. Dissolution UV-online Automatic Detection System has excellent stability, which ensures the stability and reliability of the on-line ultraviolet test results of continuous automatic sampling.
Test cases:
Perform 10 consecutive on-line sampling (5 minutes interval) for the same concentration of sample solution and detect the absorbance (Abs) of sample solution by Dissolution UV-online Automatic Detection System.
For example, in the lower concentration sample test group, the relative standard deviation (RSD) of 10 UV-online test results was 1.2%, and the difference between the average absorbance of UV-online test group (0.2916) and manual sampling control group (0.2904) was 0.4%
The test results fully show that Dissolution UV-online Automatic Detection System has excellent accuracy and stability.
Avoiding Bubble InterferenceWhen there are bubbles in the flow cell, the test results will be greatly affected. With the increase of surfactant content in dissolution medium, the probability of bubble formation and the difficulty of bubble removal will increase significantly. Thanks to the advanced sampling pump and program design, the Dissolution UV-online Automatic Detection System can effectively avoid the interference of bubbles on the test results. Even in the case of high concentration surfactant in the dissolution medium, the test results are still accurate and stable.
Test cases:
Sodium dodecyl sulfate (SDS) is a commonly used surfactant in dissolution media, which can effectively increase the solubility of insoluble samples, but its foaming ability is strong, and it is easy to produce a large number of bubbles. According to the Guiding Principles for the Determination and Comparison of Dissolution Curves of Ordinary Oral Solid Preparations issued by CFDA, the concentration of surfactants is recommended to increase gradually in the range of 0.01%-1.0%(W/V), and the concentration of specia varieties can be increased moderately.
In order to investigate the anti-bubble interference ability of Dissolution UV-online Automatic Detection System in extreme cases, the dissolution medium containing 2.5% and 5.0% (WN) sodiun dodecyl sulfate (SDS) was used in this test. Five consecutive on-line sampling sessions (5 minutes interval) were conducted for the same concentration of sample solution to observe the fluctuation of test results and compare with the manual sampling control group.
Sample solution containing 2.5% (WM) sodium dodecyl sulfate (SDS): The relative standard deviation (RSD) of the five test results was 0.4%, and the difference between the average absorbance of the UV-online test group (0.2816) and the manual sampling control group (0.2803) was 0.5%
Sample solution containing 5.0% (WM) sodium dodecyl sulfate (SDS): The relative standard deviation (RSD) of the five test results was 0.6%, and the difference between the average absorbance of the UV-online test group (0.4901) and the manual sampling control group (0.4930) was 0.6%
The results show that even if the content of sodium dodecyl sulfate (SDS) is as high as 5.0% (WW) in the sample solution, Dissolution UV-online Automatic Detection System still can effectively avoid the interference of bubbles on the test results.
Multichannel UV-Online Detection
Dissolution UV-online Automatic Detection System can connect 8 channels of UV-online detection pipe at the same time. Six of them are used for on-line detection of sample dissolution, one is used for blank calibration and one is reserved for reference standard test. By invoking the preset dissolution UVonline detection method in the operating system of automatic dissolution apparatus, the complete automatic dissolution experiment process of "dosing → running → sampling → detection" can be simply realized, which greatly improves the efficiency of the experiment.
Test cases:
By using the Dissolution UV-online Automatic Detection System, set the dissolution test parameters.as follow. Basket apparatus; Rotational Speed: 100RPM; Medium: 900ml purified water; Temperature: 37.0°℃; UV detection wavelength: 257nm. After automatic synchronous dosing, the system automatically sampled samples every 5 minutes and carried out UV-online detection. The concentration of sample solution was calculated by absorbance results, and further calculate the dissolution rate
The results of sample dissolution are as follows:
The UV-online detection results of different channels are in good agreement, the test result can accurately and real-time show the dissolution of samples. Raytor Dissolution UV-online Automatic Detection System makes drug dissolution test more accurate, convenient and efficient. |
